چکیده

Download pose: MicroRNAs (miRNA) are short noncoding RNAs that can play critical roles in diverse biolorocesses. They are implicated in tumorigenesis and function both as tumor suppressors and as enes. The clinical significance of miRNA expression profiles in malignant gliomas remains unclear. erimental Design: In this study, we examined the expression levels of 365 mature human miRNAs malignant gliomas, including 8 glioblastomas and 4 anaplastic astrocytomas, using TaqMan realuantitative PCR arrays. A validation study was done to corroborate a subset of the results, including sion levels of miR-196a, -196b, -21, and -15b, by analyzing 92 malignant gliomas by conventional me PCR. We modeled the relationship between the expression levels of these miRNAs and the al rate of 39 glioblastoma patients by Kaplan-Meier method and multivariate analysis. ults: Expression profiles in glioblastomas and anaplastic astrocytomas suggested that 16 miRNAs andidate markers associated with the malignant progression of gliomas. Among them, miR-196a d the most significant difference (P = 0.0038), with miR-196b also having a high significance .0371). Both miRNAs showed increased expression levels in glioblastomas relative to both anaastrocytomas and normal brains in the validation study. Furthermore, patients with high miRxpression levels showed significantly poorer survival by the Kaplan-Meier method (P = 0.0073). ariate analysis showed that miR-196 expression levels were an independent predictor of overall al in all 39 glioblastoma patients (P = 0.021; hazard ratio, 2.81). surviv Conclusions: Our results suggest that miR-196 may play a role in the malignant progression of gliomas and may be a prognostic predictor in glioblastomas. MiR-24 Promotes the Survival of Hematopoietic Cells

The microRNA, miR-24, inhibits B cell development and promotes myeloid development of hematopoietic progenitors. Differential regulation of cell survival in myeloid and lymphoid cells by miR-24 may explain how miR-24's affects hematopoietic progenitors. MiR-24 is reported to regulate apoptosis, either positively or negatively depending on cell context. However, no role for miR-24 in regulating ...

Role of an imbalanced miRNAs axis in pathogenesis of psoriasis: novel perspectives based on review of the literature

BACKGROUND Specific profile of microRNAs (miRNAs, miR) expressed in psoriasis has been identified in the past few years, while the studies on roles and molecular mechanisms of these miRNAs are still on the way. In our previous study, four specific miRNAs (miR-31, miR-203, hsa-miR-99a and miR-125b) were found to be specifically altered in psoriatic lesions. We therefore conducted a systematic lit...

Self-Limited versus Delayed Resolution of Acute Inflammation: Temporal Regulation of Pro-Resolving Mediators and MicroRNA

Mechanisms underlying delays in resolution programs of inflammation are of interest for many diseases. Here, we addressed delayed resolution of inflammation and identified specific microRNA (miR)-metabolipidomic signatures. Delayed resolution initiated by high-dose challenges decreased miR-219-5p expression along with increased leukotriene B(4) (5-fold) and decreased (~3-fold) specialized pro-r...

A Biogenesis Step Upstream of Microprocessor Controls miR-17∼92 Expression

The precise control of miR-17∼92 microRNA (miRNA) is essential for normal development, and overexpression of certain miRNAs from this cluster is oncogenic. Here, we find that the relative expression of the six miRNAs processed from the primary (pri-miR-17∼92) transcript is dynamically regulated during embryonic stem cell (ESC) differentiation. Pri-miR-17∼92 is processed to a biogenesis intermed...

Dual Anti-Inflammatory and Anti-Angiogenic Action of miR-15a in Diabetic Retinopathy

Activation of pro-inflammatory and pro-angiogenic pathways in the retina and the bone marrow contributes to pathogenesis of diabetic retinopathy. We identified miR-15a as key regulator of both pro-inflammatory and pro-angiogenic pathways through direct binding and inhibition of the central enzyme in the sphingolipid metabolism, ASM, and the pro-angiogenic growth factor, VEGF-A. miR-15a was down...
